1. INTRODUCTION Topical delivery can be defined as the application of a drug containing formulation to the skin to directly treat cutaneous disorders (e.g. acne) or the cutaneous manifestations of a general disease (e.g. psoriasis) with the intent of containing the pharmacological or other effect of the drug to the surface of the skin or within the skin. Semi-solid formulation in all their diversity dominate the system for topical delivery, but foams, spray, medicated powders, solution, and even medicated adhesive systems are in use. Topical delivery includes two basic types of product: i. External topical that are spread, sprayed, or otherwise dispersed on to cutaneous tissues to cover the affected area. ii. Internal topical that are applied to the mucous membrane orally, vaginally or anorectal tissues for local activity. Organogels have an organic solvent as the liquid continuous medium. Organogels are formed by specific kind of small organic molecules, which in many solvents very effectively self-assemble into a three dimensional network of nanoscale dimension, thereby turning a liquid into a gel. Organogel formation is based on the spontaneous selfassociation of individual gelator molecules into three-dimensional networks of randomly entangled fiber-like structures, thereby confining the solvent at the microscopic level. Organogels can be distinguished from hydrogels by their predominantly organic continuous phase and can then be further subdivided based on the nature of the gelling molecule: polymeric or low molecular weight (LMW) organogelators. Polymers immobilize the organic solvent by forming a network of either cross-li nked or entangled chains for chemical and physical gels, respectively. The latter is possibly further stabilized by weak inter-chain interactions such as hydrogen bonding, van der Waals forces, and π-stacking [1,2]. Advantages of organogels: i. Organogels are thermoreversible and offer improved microbial resistance as compared to aqueous solutions of hydrogels. ii. Also the network structure formed in organogels is highly ordered at the supramolecular level iii. Improved drug permeability across the skin iv. Ease of preparation and administration [3]. Organogels in drug delivery Despite the large abundance and variety of organogel systems, relatively few have current applications in drug delivery, owing mostly to the lack of information on the biocompatibility and toxicity of organogelator molecules and their degradation products. Bifonazole is a novel imidazole antifungal drug which has been used in many skin and nail infections caused by fungi. Although, topical formulation of the drug is available but, to investigate the potential of organogel as a novel topical drug delivery system, the above drug candidate was selected. The objective of the present study is Development of a novel topical vehicle for the delivery of an antifungal agent in terms of effectiveness and elegance as compared to conventional ointment and creams, improved availability at the desired site by use of organogels and Improved patient compliance [4,5].
2. INTRODUCTION Gels: A gel is a solid semi-solid system of at least two constituents, consisting of a condensed mass enclosing and interpenetrated by a liquid. When the coherent matrix is rich in liquid, the product is oft en called as jelly. The United States pharmacopoeia defines gel as “semisolid, being other suspension of small inorganic particle or large inorganic molecules interpenetrated with liquid”. This is a true phase system, as an inorganic particle is not soluble but merely dispersed throughout the continuous phase. 1
CLASSIFICATION OF GELS 1. On the basis of phase system Two phase system: The gel mass may consist of floccules of small particles rather than large molecules. The gel structure in these systems is not always stable. Such gels may be thixotropic, forming semisolid on standing and become liquid on agitation. e.g. aluminium hydroxide gel, bentonite magma. Single phase system 2. On the basis of Chemical Nature Inorganic gel Organic gel 3. On the basis of structural arrangement Physical gels: unstable at high temperatures or in solvents. Chemical gels: not tough due to uneven structure. Slide-ring gels: these gels are differ from physical and chemical gels in that the polymer chains are neither

NEED OF ORGANOGELS The organogel do not form semisolids on standing. Because an organogel may consists of macromolecules existing as twisted matted strands. The units are of bound together by strong types of Vanderwaal forces so as to form crystalline amorphous regions throughout the entire system. 4,5 The organogels have lower hydrations, the drug dissolving polymer and is transported between the chains. Cross linking increases hydrophobicity of gels & diminishes the diffusion rate of drug. 6 Advantages: diffusion rate 0f drug. Because the drug is dissolved in polymer & transported between chains. Disadvantages: syneresis. When the gel is taken up of liquid with an increasing volume known as swelling.
ORGANOGEL STRUCTURE & MECHANISM OF ORGANOGELLING The organogelling or the gelation of the lecithin solutions in organic solvents is induced as a result of the incorporation of a polar solvent. Approaches developed by Israelachvili et al, the aggregate transformation (i.e. sphere-to-cylinder transformations) are determined by a change of a curvature for the amphipmile monolayer.7 In particular, the effects of polar solvent introduced into spherical lecithin micelles may be associated with an increase in the cross-sectional area of the lecithin polar region in which the solvent is arranged. The shape of the hydrated molecules is close to a cylinder. This shape leads to packing constraints in the s pherical micelles that are diminished through the transition into the cylindrical ones with a smaller curvature.
PENDAHULUAN Pengiriman topikal dapat didefinisikan sebagai penerapan obat yang mengandung formulasi pada kulit untuk mengobati gangguan kulit secara langsung ( misalnya jerawat ) atau kulit manifestasi dari penyakit umum (misalnya psoriasis ) dengan maksud mengandung efek farmakologis atau lainnya obat ke permukaan kulit atau dalam kulit . Formulasi semi- padat dalam segala kea nekaragamannya mendominasi sistem untuk pengiriman topikal , tapi busa , semprotan , bubuk o bat , solusi , dan sistem perekat bahkan obat yang digunakan . Pengiriman topikal mencakup dua tipe dasar produk : i . Topikal eksternal yang tersebar , disemprot , atau tersebar pada jaringan kulit untuk menutupi daerah yang terkena . ii . Topikal internal yang diterapkan pada membran mukosa oral , vagina atau jaringan anorectal untuk kegiatan lokal . Organogels memiliki pelarut organik sebagai media terus menerus cair. Organogels dibentuk oleh jenis tertentu dari molekul organik kecil , yang dalam banyak pelarut sangat efektif merakit diri ke dalam jaringan tiga dimensi dari dimensi nano , sehingga mengubah cairan menjadi gel . Formasi Organogel didasarkan pada self- asosiasi spontan molekul gelator individu menjadi jaringan tiga dimensi secara acak terjerat seperti serat struktur , sehingga membatasi pelarut pada tingkat mikroskopis . Organogels dapat dibedakan dari hidrogel oleh fasa kontinyu terutama organik mereka dan kemudian dapat dibagi lagi berdasarkan sifat molekul pembentuk gel : berat molekul polimer atau rendah ( LMW ) organogelators . Polimer melumpuhkan pelarut organik dengan membentuk jaringan baik cross-linked atau tersangkut rantai untuk gel kimia dan fisik , masingmasing. Yang terakhir ini mungkin lebih stabil oleh lemahnya interaksi antar - rantai seperti ikatan hidrogen , gaya van der Waals , dan π - stacking [ 1,2 ] . Keuntungan dari organogels : i . Organogels yang termoreversibel dan menawarkan peningkatan resistensi mikroba dibandingkan dengan larutan berair dari hidrogel . ii . Juga struktur jaringan yang terbentuk di organogels sangat memerintahkan pada supramolekul tingkat iii . Peningkatan permeabilitas obat melintasi iv kulit . Kemudahan persiapan dan administrasi [ 3 ] . Organogels dalam pemberian obat Meskipun kelimpahan besar dan berbagai sistem organogel , relatif sedikit memiliki aplikasi saat ini dalam pemberian obat , karena sebagian besar kurangnya informasi tentang biokompatibilitas dan toksisitas molekul organogelator dan produk degradasi mereka . Bifonazole adalah obat antijamur imidazol baru yang telah digunakan di banyak infeksi kulit dan kuku yang disebabkan oleh jamur . Meskipun , formulasi topikal obat ini tersedia tetapi , untuk menyelidiki potensi organogel sebagai sistem pengiriman obat topikal baru , calon obat di atas dipilih . Tujuan dari penelitian ini adalah Pengembangan kendaraan topikal baru untuk pengiriman agen antijamur dalam hal efektivitas dan keanggunan dibandingkan dengan salep dan krim konvensional , peningkatan ketersediaan di situs yang diinginkan dengan menggunakan organogels dan Peningkatan kepatuhan pasien [ 4 , 5 ] .

Amerika Serikat farmakope mendefinisikan gel sebagai " semipadat , menjadi suspensi lain dari partikel anorganik yang kecil atau molekul anorganik besar interpenetrated dengan cairan " . Organogel STRUKTUR & MEKANISME ORGANOGELLING The organogelling atau gelasi dari solusi l esitin dalam pelarut organik diinduksi sebagai hasil dari penggabungan pelarut polar . Pendekatan yang dikembangkan oleh Israelachvili et al , transformasi agregat ( yaitu transformasi bola -to - silinder ) ditentukan oleh perubahan kelengkungan untuk monolayer.7 amphipmile Secara khusus, efek dari pelarut polar diperkenalkan ke misel lesitin bola dapat dikaitkan dengan peningkatan luas penampang dari daerah kutub lesitin di mana pelarut diatur . Bentuk molekul terhidrasi dekat dengan silinder . Bentuk ini menyebabkan kendala packing dalam misel bola yang berkurang melalui transisi ke yang silinder dengan kelengkungan yang lebih kecil .
